The article presents the results of the floristic study in the lower reaches of the Sukhona River basin (within the limits of Babushkinsky, Velikoustyugsky, Nyuksensky, Tarnogsky and Totemsky districts of the Vologda Region) conducted as a part of the regional Red Data Book maintenance. Field surveys of 2004-2022 allowed to identify a total of 753 new localities of 67 protected and 47 rare species of vascular plants. Populations of two plant species listed in the Red Data Book of the Russian Federation ( Cypripedium calceolus , Orchis militaris ) were discovered. The most significant findings also include the occurrences of Adonis sibirica , Anemonoides altaica , Corydalis capnoides , Geranium robertianum , Gymnadenia densiflora , Hedysarum alpinum , Onobrychis arenaria , Spiraea media . New records of 41 protected species and 29 species that require scientific monitoring in the region, are provided for seven regional specially protected natural areas (geological reserve (zakaznik) “Urochishche “Strel'na”, landscape reserves (zakaznik's) “Pin'ga” and “Ramenskiy les”, nature rezervat “Volguzh”, and natural monuments “Geologicheskoye obnazheniye u dereven' Purtovino i Isady”, “Mys Byk”, “Opoki”). Less than 40% of the analyzed species were recorded outside the boundaries of protected natural areas.
