An improved method of purification of potato potyvirus A (PVA) has been developed. The method enables to obtain as much as 2 mg of highly purified virus preparation from 40 g of Nico-tiana clevelandii fresh leaves. The monospecific rabbit antiserum with high liter to the PVA has been prepared and double antibody sandwich method of enzyme-linked immunosorbent assay (das-ELISA) has been developed for the detection of the PVA. The sensitivity of the horse-radish peroxi-dase-conjugated das-ELISA was about 8-16 ng/ml permitting to detect PVA in seed potatoes.
