Clonal reproduction in vitro of flowering raspberry

Бесплатный доступ

The mineral, carbohydrate and hormonal compositions of nutrient media for in vitro reproduction of flowering raspberry have been optimized. It was found that the use of nutrient media according to the prescriptions of QL and Anderson allows to obtain higher reproduction coefficients than the use of the MS medium, traditional for plants of the genus Rubus. The best result (reproduction coefficient 1.76, average shoot length 18 mm) was recorded on MS medium with half concentrations of nitrogen, phosphorus, potassium and magnesium. The use of thidiazuron (TDZ) in concentrations of 0.1-0.2 mg/l in the process of reproduction of flowering raspberry in vitro allowed to obtain multiplication coefficients by 37-67.7% more than when using 6-BAP. TDZ causes a slight decrease in the average length of the shoots formed. Therefore, at the multiplication stage preceding rooting, it should be replaced with 6-BAP. As a source of carbohydrate nutrition, the most effective use of sucrose or glucose in concentrations equal to 20 g/l.
