Метаболиты базидиальных грибов, эффективные в терапии рака и их молекулярные мишени: обзор

Рассмотрены история и современное состояние использования продуцируемых грибами базидио-мицетами веществ в терапии рака. Перечислены молекулярные мишени рака, связанные с его базовыми аспектами - митогенной гиперактивностью, уклонением от апоптоза, неоангиогенезом, клональной гетерогенностью. Показано, что грибные метаболиты могут быть задействованы в успешном воздействии практически на все основные мишени рака. Подробно рассмотрено опосредованное естественными киллерными клетками онкостатическое действие грибных р-глюканов. Показаны свойства ряда грибных метаболитов (хиноны, терпеноиды) игнибировать протеинкиназы, циклин-зависимые киназы, рецепторы эпидермального и эндотелиального факторов роста, хемо-кин NF-kB и металлопротеиназы, что ведет к аресту клеточного цикла раковых клеток, торможению неоангиогенеза, разблокировке проапоптотических путей и торможению опухолевой инвазии. Упоминаются грибы - продуценты витальных ядов и способ адресной доставки токсина до раковой ткани. Обсуждаются перспективы исследований лекарственных свойств базидиальных грибов. Показана необходимость сравнительного изучения воздействия низкомолекулярных ингибиторов пролиферативных путей на дифференцированные и стволовые раковые, а также здоровые проли-ферирующие клетки в свете представлений о циклозависимой химиотерапии. Обосновывается необходимость профилирования видов грибов по соотношению иммуностимулирующий и противовоспалительной активностей. Названы новые перспективные виды, сочетающие иммуностимулирующие и противовоспалительные свойства - Taiwanofungus camphoratus и Piptoporus betulinus. Обсуждаются онкопревентивные свойства базидиальных грибов.
