Two methods of chromatographic purification for manufacture of preparation of human coagulation factor VIII at a dosage of 500 IU/vial were compared. Size exclusion chromatography and ion-exchange chromatography were used in method 1, ion-exchange chromatography only was used in method 2. It was found that despite lower recovery, method 1 provided higher level of purification of factor VIII in comparison with method 2. Thus, purification method 1 is preferable for preparation of human coagulation factor VIII.
