Stabilization of the monogermity trait when developing sugar beet hybrid components

Development of genetical monogerm sugar beet hybrids is of great importance for farmers as it is a prerequisite for introduction of technologies of sugar beet industrial cultivation without manual labour inputs. It can be achieved if factory seed lots have minimum monogermity and high germinating capacity being the result of periodic selections and evaluation using monogermity loci. The method of unfertilized ovules induction and cultivation of haploid regenerants that are pollinators - sterility fixers has proved to be effective. Efficiency of RFLP-analysis method using Hind III restrictase that has allowed identifications of haploid microclones according cytoplasm type is shown. Molecular markers have indicated that regenerants with normal cytoplasm (N) have one PCR-product of 800 bp in length; two products of restriction (320 bp and 480 bp) have been found in the forms (S) with cytoplasmic male sterility (CMS). Detection of haploid regenerants with sterile cytoplasm from initial population is of great theoretical and practical importance for breeding thus facilitating the problem of producing homozygous lines with CMS and high-productive hybrids on the sterile basis. Final processes of the technology are: transfer of rooted microclones with a vigorous root system and well developed leaf area to greenhouse conditions, growing seed-bearing plants from them, and obtaining of seeds of DH-lines.
