The effect of mononuclear fraction of autologous bone marrow and of the drug “Myelopid” on the microcirculation in critical limb ischemia in experiment

The experiments were conducted on 100 white male Wistar rats, which were divided into four groups: intact, control, experimental and comparison group. In the intact group consisted of 10 rats, which evaluated normal values of microcirculation. In the other groups, 30 animals were included with modeling of critical limb ischemia. In the control group, treatment was not carried out. In the comparison group, laboratory animals obtained mononuclear fraction of autologous bone marrow according to the original method. In the experimental group, the treatment was carried out in a combined way: mononuclear fraction of autologous bone marrow and the drug "Myelopid”. The assessment of the level of perfusion in the muscles of the lower leg of rats was performed by laser Doppler flowmetry on the 10th, 21st and 28th days. At the same time made a histological examination of the ischemic muscle definition level dynamics arterio-venular shunting, the area of necrosis and the density of the capillary network. The combined use of mononuclear fraction of autologous bone marrow and immunostimulating drug "Myelopid” in the treatment of experimental critical limb ischemia to a greater extent increases the level of soft tissue perfusion and capillary network density, which leads to a decrease in the index of arterio-venular shunting and the area of necrotic tissues, than with isolated administration of bone marrow cells.
