Influence of acute gamma irradiation on cell cycle parameters in 293 HEK cell culture

Purpose. The aim of the work was to determine change of the quantitative indicators depending on the cell cycle after one fraction gamma irradiation in vitro. Materials and methods. Cells (line 293 HEK) got single fraction irradiation on gamma-ray equipment "ROKUS-AM". Cell cycle was analyzed on Beckman Culter FC 500 flow cytometer. Standard PI-based method of dyeing was used. The main results. Quantitative and qualitative assessment of radiosensitivity and changes in parameters of cell cycle after a single exposure to a dose of 2 to 10 Gy was carried out. Cell death after dose of 10 Gy reached 40%. After irradiation cell stopped proliferation at the G2M phase. This effect depended on dose and 90% cells stopped at G2M phase 24 hours after 10 Gy. After 48 hours most cells still were resting in G2M phase of cell cycle.
