New food peptide cell photoaging for preventing: perfomans and in vitro confirmation of effectiveness

The peptide was designed on the Enalos InSilicoNano platform using the PubChem database as part of virtual peptide creation and screening. A peptide with the molecular formula C83H150N24O24S4 and the sequence of amino acids was obtained: CTKSICTKKTLRTCPPIC. Its use as a substance with the necessary physico-chemical properties for effective use in food products and as an active ingredient in appropriate biologically active additives is shown. The total hydrophobic ratio determined by APD is 39%, the hydrophobicity of the peptide according to Wimley-White in the whole residue is 3.23. The binding potential with proteins (Boman index) is: 1.4 kcal / mol. The external extinction coefficient is 250 М-1 sм-1 at wavelength of 280 nm measured in water. Estimated in vivo half-life is more than 10 hours. Instability index of the studied peptide is 37.77. Index of hydropathy of the studied peptide is -0.011. It was shown that the treatment of human lung fibroblast cells (Fibr) with ultraviolet radiation significantly (p≤0.01) reduced the viability of cells by 47%, the cultivation of cells with the peptide significantly reduced negative effects of ultraviolet radiation. Thus, the viability of fibroblasts when calcined with the peptide and irradiated with ultraviolet rays was 92%. Accordingly, use of the peptide reduces the effect of photoaging cells by 1.9 times. It has been established that the studied peptide is unique, non-toxic and non-antigenic. In an in vitro experiment, it was shown that the resulting peptide prevents photoaging of cells.
