The optimization of DNA extraction of broomrape (Orobanche cumana Wallr.) and the identification of polymorphic RAPD and SSR-loci for genotyping of parasite

The methodology for DNA extraction from the fresh and dried-up tissues of broomrape (Orobanche cumana Wallr.) plants at different stages of ontogenesis was optimized. The DNA extraction from the fresh frozen inflorescences by the method of Doyle J.J. and J.L. Doyle (1987) with an addition of 1 % of polyvinyl pyrrolidone to the extraction buffer was considered the optimum method of extraction. Three RAPD-and ten SSR-primers that initiate amplification of polymorphic fragments of DNA were selected. There were identified the steadily reproducing fragments of amplificated DNA which can serve as reliable markers for studying of inter- and intra-population variability of parasite populations of the South of Russia.
