Acute toxicity and cumulative properties of the local anesthetic ropivacaine hydrochloride (Naropin®)

Naropin ® - a new amide local anesthetic with long duration of action. Naropin ® is still not intensively used in veterinary practice, so the purpose of this study is to provide accurate data about the parameters of acute toxicity and cumulative effect of a 1% Naropin ® in rats. It was found that the LD50 of the intraperitoneally injected Naropin® in rats was 72,0 ± 4.8 mg / kg versus 75,5 ± 2.8 mg / kg in female white rats during subcutaneous administration. Index of cumulation (I cum) during acute experiment was 0. Seizures followed by depression are the main signs of acute intoxication of the drug. Sub-chronic LD50 is 677.7 mg / kg for white female rats, and the coefficient of cumulation is 8,97. Finally, Naropin ® has a very narrow zone of toxicity with a mild type of cumulation during the single and long-term use.
