A method for obtaining brucellosis serum for diagnostic studies in brucellosis of animals

This article describes a method for obtaining hyperimmune Brucella serum, taking into account the recommendations and requirements of the OIE. An immunization scheme for bulls was developed, the brucellosis strain Brucella abortus 544 was used as an antigen, cultural-morphological, tinctorial and agglutinogenic properties were studied, which corresponded to the passport data of the strain. The inactivated antigen was administered 3 times at a dose of 20.0 cm3, with an interval of 7 days. Blood samples were taken 7-10 days after the last antigen injection. After that, the serum was separated and tested for activity and specificity by setting the complement fixation reaction (CFR) and the agglutination reaction (RA) to the limiting titer with the brucellosis antigen. The proposed method for obtaining hyperimmune Brucella serum for serological reaction provides for the production of highly active and highly specific agglutinating S-serum with limiting titers in RSK 1:160 and in RA 1:1600 in a short time. The resulting brucella serum is applicable as a national standard and as a specificity control in the study of sera with R- and S-antigens and the activity of S-antigens, as well as in the study of the dissociation of industrial Brucella strains used for the manufacture of vaccines and laboratory strains during research works.
