Association of polymorphism 5G/4G PAI-1 gene with the developmentof thrombophilic states in chronic myeloproliferative diseases

Studying the association of thepolymorphism of the PAI-1 gene with thedevelopment of thrombophilic conditions inchronic myeloproliferative diseases. The workwas performed on DNA samples isolated fromthe peripheral blood of patients with the mostcommon chronic myeloid leukemia (CML,n = 32) and polycythemia vera (PV, n = 79), aswell as the control group (n = 114). Genotypingof the samples and detection of the 5G/4Gpolymorphism of the PAI-1 gene were performedby PCR on an “Applied Biosystems 2720” device(USA) using the “Litekh” reagent kits (Moscow,Russia). Statistical analysis of the resultswas performed using the statistical softwarepackage “OpenEpi 2009”. Results and discussion.Heterozygous and homozygous genotypes of the5G/4G polymorphism of the PAI-1 gene in thesubgroup with thrombosis in groups of patientswith CML and PV increases the risk of thrombosis.Perhaps this is due to the fact that, in individualswith a heterozygous genotype, the protectiveeffect of the wild genotype 5G/5G is lost, as aresult of which thrombophilic conditions maydevelop.
