Investigation of the efficacy of different variants of chimeric T-cell receptors for CEA-positive tumor cells

Summary Oncological diseases are one of the leading causes of mortality worldwide. Immune cells affect the development of malignant tumors. Methods of cancer therapy based on modification and activation of own immune cells have been developed, so that they can selectively affect only tumor cells. Cancer embryonic antigen (CEA) is one of the most common and universal markers for most tumors. We modified lymphocytes with PCI-1C6-CAR and PCI-3C1-CAR-new plasmids carrying the coding sequences of various T-cell receptor antigens. Then the cytotoxic activity of the modified lymphocytes against CEA-positive tumor cells was studied. It was shown that these lymphocytes selectively affect cells expressing CEA. A comparative analysis of antitumor activity of lymphocytes transfected with various plasmids was also performed. In cell cultures A549 and HST116, the highest cytotoxic activity of lymphocytes transfected with plasmid PCI-3C1- CAR-new was revealed.
