Assessment of the influence of the germination process on store proteins (SDS-PAGE) and preservation of genomic DNA of Triticum Aestivum L.

The paper presents a comprehensive comparative analysis of the degradation of gluten storage proteins and the integrity of genomic DNA in intact and sprouted grain of spring soft wheat (Triticum aestivum L.). The relevance of the study is due to the fact that grain germina-tion in the field, due to unstable climatic conditions observed over the past several years, leads to the activation of endogenous hydrolases and a significant decrease in grain quality. However, the effect of this process on the integrity of genomic DNA has not been sufficiently studied. Protein analysis was performed using vertical SDS-PAGE under non-reducing conditions on a 10 % poly-acrylamide gel with a 10–180 kDa marker. Genomic DNA was isolated using the HiPurA SuperPlant DNA Purification Kit (HiMedia, India) and analyzed by electrophoresis in a 1% agarose gel and spectrophotometrically (A260/A280). It was established that sprouted wheat grains exhibit significant degradation of high-molecular-weight glutenins (HMW-GS, 100–130 kDa) and a decrease in the intensity of LMW-GS and γ-gliadin bands (35–50 kDa) with the accu-mulation of low-molecular-weight proteolysis products (< 15 kDa). Genomic DNA analysis showed that high-molecular-weight fragments retain their integrity in all samples, including sprouted ones. However, the DNA concentration in sprouted samples (0.10 mg/ml) is lower than in intact ones (0.11–0.13 mg/ml), and the purity index A260/A280 = 1.65 indicates contamination with protein impurities, requiring additional purification. It was concluded that at the early stages of germination, the activation of proteases outpaces the activation of nucleases, as a result of which the sprouted grain is suitable for molecular genetic identification and genotyping of wheat varieties.
