Synthesis of complex D-mannitol (E 421)-based additives to increase bioavailability of nitrogenous bases

Currently, one of the most important areas of food supplement and drug form industries is the production of complex additives based on biologically active components. One of the most important parameters of such systems is increased rate of bioactive substance release in solutions, which determines high bioavailability of active substance. The paper presents research results of synthesis, identification, and study of dissolution dynamics of D-mannitol (E 421) molecular complex with adenine. The synthesis was carried out using a partially deprotonated form of adenine (adeninium-adenine hydrosulphate), which had been obtained previously. The complex structure study showed participation of free amino group in binding to secondary hydroxyl groups of mannitol via hydrogen bonds, probably with partial proton transfer of hydroxyl groups. The study of complex dissolution dynamics in comparison with adenine shows a noticeable increase in release of nitrogenous base in the first case, which is probably due to modification of relatively hydrophobic adenine structure by D-mannitol hydrophilic molecules.
